The course was designed by Pete Dye in 1967 with the help of Jack Nicklaus. Dye also designed another course in the Sea Pines Resort, Heron Point.

Harbour Town Golf Links is a par 71 course and 7,099 yards (6,491 m) from the back tees which is relatively short for a PGA Tour event.

The course is famous for its finishing holes, particularly the par-4 18th, which is one of the most recognizable holes in golf. The hole runs along the Calibogue Sound and features the iconic Harbour Town Lighthouse in the background, making it a picturesque and challenging finishing hole.

Harbour Town is known for its narrow fairways, small greens, and strategic bunkering. The course emphasizes accuracy and shot-making over power, requiring players to carefully consider each shot.

Since 1969, it has hosted the RBC Heritage which is played mid-April, the week after The Masters. The tournament is one of the most beloved stops on the PGA Tour, attracting top players and large crowds due to its unique atmosphere and challenging course.

The inaugural tournament, "The Heritage Golf Classic", was played in late November 1969. The winner's share was $20,000.
